Hypericum sinaicum L. is an endangered Egyptian medicinal plant of high importance due to the presence of naphthodianthrones (hypericins), which have photodynamic properties and pharmaceutical potential. We sought to assess H. sinaicum ability to develop hairy roots that could be cultured in contained conditions in vitro and used as a source for hypericin production. We used four A. rhizogenes strains differing in their plasmids and chromosomal backgrounds to inoculate excised H. sinaicum root, stem and leaf explants to induce hairy root development. Anatomical and histochemical aspects of zigotic embryo and leaves in 'Coqueiro Anão' Ciência Rural

The paper provides information about histochemical staining reactions in leaves and embryos of 'Coqueiro Anão' (Cocos nucifera). It was compared in vitro coconut and autotrophic palm leaves. Reactions for insoluble polysaccharides and acidic compounds, protein, extractable lipids, lignin and other classes of compounds were tested using histochemical tests. None sample gave positive reaction for lignin and phenolic compounds. All the samples gave positive reaction for protein, starch and insoluble polysaccharides while acidic compounds were positive only in in vitro leaves. ABSTRACT This study developed a methodology for callus induction in leaf segments of B. verbascifolia and evaluated the bioactive phenolic compounds production. Leaf explants were cultured in MS medium with 30 g L-1 sucrose, solidified with 7 g L- 1 agar supplemented with 2,4-D (0; 4.52; 9.05; 18.10 µM) and BAP (0; 4.44; 8.88; 17.75 µM ) in the presence and absence of light. ABSTRACT: The aim of the present study was to evaluate the effects of carbohydrate supplementation on the propagation of the orchid Cattleya schilleriana. The 120-d-old seedlings were subcultured in fructose-, glucose-, or sucrose-supplemented (0, 15, 30, and 45g L-1) ½ MS culture medium (half-strength macronutrient concentrations), using a completely random design with four repetitions per treatment. The aim of the present research was to verify the in vitro growth of orchids in different systems of micropropagation, being cultivated in a bioreactor, with natural ventilation and conventional systems. Cattleya walkeriana plants were obtained from the germination of seeds in culture medium. After 8 months, seedlings with 1 cm of length were placed in a culture vessel according to the treatments, which counted with two micropropagation systems (conventional and natural ventilation) in three media of culture (liquid, solid with 5 or 6g L-1 of agar). Epiphytic orchids have been considered an interesting model for plant development and may have unusual features related to the hormonal control of shoot and root growth. In the present experiment we have studied two genotypes of Catasetum fimbriatum Lindl. (Orchidaceae) with an inverse pattern of shoot and root dry matter accumulation. Whereas the CFC1 genotype directs 60% of dry matter towards the shoot system, this same amount is directed towards the root system in the CFC4 genotype. Differences in the endogenous content of cytokinins, IAA and ABA, as well as differences in IAA conjugation and oxidation in shoots and roots were observed in the two genotypes. In vitro techniques comprise valuable tools for the conservation of endangered plant species. The aim of the present study was to evaluate the morphological features, global DNA methylation levels and free polyamines during protocorm-like bodies (PLBs) induction of C. tigrina. Ovaries were collected from adult cows and fragments of ovarian cortex were immediately fixed (non-cultured control) or cultured in vitro in α-MEM+ alone or containing 10, 50, 100 or 1,000ng/mL rbST. Effect of supplementation of green tea polyphenols on the developmental competence of bovine oocytes in vitro BJMBR

The objective of the present study was to examine the effect of green tea polyphenols (GTPs) supplementation during in vitro maturation, in vitro fertilization, and in vitro culture on the developmental competence of bovine oocytes. Cumulus-oocyte complexes aspirated from the ovaries were matured in vitro (38.5ºC for 24 h) and fertilized (38.5ºC for 15-18 h) and embryos were cultured (38.5ºC for 192 h) in a defined conditioned medium with or without GTPs supplementation. The effect of inoculum size on the axenic in vitro production of Steinernema carpocapsae, using a transparent medium with hemoglobin and cholesterol, was studied. Axenic cultivation was performed with fertile eggs ofS. carpocapsae (hatching of eggs: 89 ± 4%), which were obtained by lyses of female nematodes and subsequent incubation. Two important aspects of this work include the use of a completely transparent medium enriched with hemoglobin and cholesterol and the high multiplication factor of the population (MFIP) of 87, which is considered high for in vitro axenic cultures of nematodes. An effective tissue culture is an indispensable basis for the production of transgenic plants. In this study, we investigated the induction of embryogenic callus production from the scutellum of spring wheat and subsequent regeneration of plants. Twenty-two Argentine spring wheat varieties from three breeders were screened to select promising genotypes for the optimization of the procedure. Experimental variables were the developmental stage of the immature embryos and the concentration of the growth regulator 2,4-dichlorophenoxyacetic acid (2,4-D). The interspecies germ cell transfer technique has been suggested as a way to conserve endangered birds.
